Jose made her debut television appearance on ABS-CBN. She played Esther Lagrimas in the hit series Familia Zaragoza from 1995 to 1996. From 1997 to 1999, Jose played the role of Magda in the original 1997 series Mula Sa Puso, before making her first appearance on Maalaala Mo Kaya.

Throughout her career, Jaclyn Jose has acted in many movies and shows on two stations, ABS-CBN and GMA Network, including Hiram (2004-2005), Komiks Presents: Agua Bendita (2006), Love Spell (2007-2008), Zorro (2009), Felix Manalo (2015), Ma’ Rosa (2016), Mang Kepweng: Ang Lihim ng Bandanang Itim (2020), etc.

Especially in 2016, she won the “Best Actress” award at the Cannes Film Festival for her role as a mother in the film Ma’ Rosa. This is the first time a Filipino and Southeast Asian actress has achieved this title.

Ma’ Rosa is about a mum-of-four who owns a small convenience store in a poor neighbourhood of Manila. To make ends meet, she and her husband resell small amounts of illegal drugs on the side. When they get arrested, she and her children become desperate and will do anything to buy her freedom from corrupt police officers.

The movie was directed by Brillante Mendoza, who in 2009 became the first Filipino to win best director at the festival for Kinatay.
